We examined cluster root formation and root exudation by white lupin (Lupinus albus L. cv. Kiev Mutant) in response to growth medium and phosphorus supply in a sand/solution split-root system. The split-root
system consisted of a nutrient solution compartment and a siliceous sand compartment. Phosphorus was applied at 1 (low-P plants)
or 50 (high-P plants) μM as KH2PO4 to the solution compartment and at 10, 50 or 250 mg P kg−1 as hydroxyapatite (Ca-P) to the sand compartment. In contrast to the high-P plants, P concentration and P uptake in the low-P
plants increased with increasing P supply to the sand compartment. The NaHCO3-extractable P was lower in the rhizosphere of the low-P plants than the high-P ones. The proton extrusion rate by the solution-grown
roots of the low-P plants was higher than that of the high-P plants at the early growth stage. For the low-P plants, the proportion
of dry root biomass allocated to cluster roots was higher in the solution compartment than that in the sand compartment. The
citrate exudation increased in the sand compartment and decreased in the solution compartment with time, showing a lack of
synchronization in citrate exudation by two root halves grown in different media. The cluster root proportion and citrate
exudation in both compartments decreased with increasing shoot P concentration. An additional experiment with no P added to
either root compartment showed that the proportion of cluster roots was about 9% lower in the sand than solution compartments.
The results suggest that cluster root formation and citrate exudation can be significantly affected by the root growth medium
in addition to being regulated by shoot P status. More P can be exploited from sparingly available Ca-P by the low-P plants
than the high-P ones due to greater citrate exudation under P deficiency. 相似文献

构建含Endostatin基因的腺病毒载体,将Endostatin基因导入培养的角朊细胞,并采用套皿法共培养角朊细胞与内皮细胞,测定培养液中Endostatin含量、内皮细胞增殖周期各时相比例、内皮细胞凋亡及细胞抑制率。结果表明转染Endostatin基因的角朊细胞可有效表达并分泌Endostatin,连续培养3天后,培养液中Endostatin含量可达226ng/ml;与转基因角朊细胞共培养的内皮细胞凋亡百分数与抑制率分别为(32.7±7.1)%、(60.5±8.3)%,均显著高于对照组[(7.3±2.0)%,(13.8±1.6)%],且G0/G1期比例明显高于对照组,而S期、G2/M期比例及增殖指数显著低于对照组。因此,转染Endostatin基因角朊细胞与内皮细胞共培养时,角朊细胞可通过分泌Endostatin促进内皮细胞凋亡,并抑制其增殖。 相似文献

氧化修饰是细胞内的活性氧诱导生物大分子发生氧化反应引起的结构及构象改变,发挥调控信号转导和对应激作出反应的功能。氧化修饰发生在凋亡信号转导中的多个生物大分子,包括凋亡相关蛋白质的氧化,如caspase-9、线粒体通透性转变孔及电压依赖的阴离子通道(voltagedependent anion channel,VDAC),同时也包括膜磷脂的氧化修饰,如磷脂酰丝氨酸及线粒体特异的心磷脂。氧化修饰作用也涉及凋亡诱导因子、促凋亡的凋亡信号调控激酶1(apoptosis signalregulatin gkinasel,ASK1)信号转导途径及抗凋亡的转录因子NF—kB的激活和活性。所以氧化修饰可能是调控凋亡信号转导机制中除磷酸化、泛素化外的另一个新的分子机制。 相似文献

钠钙交换是小鼠心脏发育中最早有功能性表达的通道基因。它的功能主要是通过泵出1个钙,泵入3个钠位置细胞内的钙稳态,此外可能参与兴奋收缩偶联。但是,至今钠钙交换在心脏发育过程中的功能性表达及其在细胞早期兴奋形成中的作用还不是很清楚。采用胚胎干细胞分化的心肌细胞为研究对象,发现在发育极早期,电压钳制在35mV的条件下,10mmol/L咖啡因诱导的内向电流的80%能被灌流液中Na^+被等浓度的Li^+取代(n=8)。此为钠钙交换电流。所有钳制的细胞单细胞RT-PCR都检测到了NCX1亚型的mRNA表达。进一步研究了钠钙交换的功能,发现等浓度Li^+取代灌流液中Na^+及应用高浓度Ni^2+阻断了膜电位震荡及与震荡相间的动作电位(早期膜兴奋形式)。因此认为钠钙交换(NCX1亚型)在心脏发育极早期的心肌细胞中已有大量功能性表达,它对于早期自主性兴奋活动的发生起着关键性的作用。 相似文献

污水处理厂是抗生素抗性基因(antibiotic resistance genes,ARGs)和抗生素抗性细菌(antibiotic resistant bacteria,ARB)重要的源和汇,生物气溶胶是ARGs和ARB自污水处理厂向周边环境释放的关键载体。目前缺乏对污水处理厂生物气溶胶抗生素抗性污染特征、来源及潜在风险的系统性总结。本文从采样方法、检测方法、逸散特征、来源、潜在危害和风险评估等方面对污水处理厂抗生素抗性污染研究现状进行综述。惯性采样法和过滤法是常用的污水处理厂抗生素抗性生物气溶胶主要采集方法,而宏基因组测序、组装和分箱为其ARGs组成、可移动性和宿主提供了有效的检测方法,抗多药类、抗杆菌肽类、抗氨基糖苷类、抗四环素类、抗β-内酰胺类、抗磺胺类、抗大环内酯类和抗糖肽类等抗性基因在污水处理厂PM10、PM2.5和PM1.0颗粒物中广泛检出。格栅间、生化反应池和污泥处理单元是污水处理厂PM10、PM2.5和PM1.0负载ARGs和ARB的主要释放单元。污水处理厂不同粒径生物气溶胶中致病性ARB的存在增加了抗生素治疗的难度,而污水和污泥对ARGs和ARB的释放起到了重要的源的贡献。本文在研究内容、研究技术和控制策略等方面也提出了相关展望,以期为污水厂生物气溶胶抗生素抗性污染的监测和防护提供参考和借鉴。 相似文献

In this paper we report on the molecular cloning, sequencing and partially characterisation of a lectin from bulb of the Chinese
medicinal plant Zephyranthes grandiflora. The full-length cDNA of Z. grandiflora bulb lectin (ZGBL) consisted of 986 bp and contained a 576 bp ORF encoding a 191 amino acid protein. Bioinformatics analysis
results clearly indicate that ZGBL belongs to the monocot mannose-binding lectin family, which contains 3 putative mannose-binding
sites per subunit. RT-PCR analysis results indicate that ZGBL is constitutively expressed in all the tested tissue types including root, bulb, leaf and flower. Interestingly, ZGBL is

5-enolpyruvylshikimate 3-phosphate synthase (EPSPS; 3-phosphoshikimate 1-carboxyvinyl-transferase; EC 2.5.1.19) is a critical
enzyme in the shikimate pathway. The full-length EPSPS cDNA sequence (CaEPSPS, GenBank accession number: AY639815) was cloned and characterized for the first time from woody plant, Camptotheca acuminata, using rapid amplification of cDNA ends (RACE) technique. The full-length cDNA of CaEPSPS was 1778 bp containing a 1557 bp ORF (open reading frame) encoding a polypeptide of 519 amino acids with a calculated molecular
mass of 55.6 kDa and an isoelectric point of 8.22. Comparative and bioinformatic analyses revealed that CaEPSPS showed extensive
homology with EPSPSs from other plant species. CaEPSPS contained two highly conserved motifs owned by plant and most bacteria
EPSPSs in its N-terminal region. Phylogenetic analysis revealed that CaEPSPS belonged to dicotyledonous plant EPSPS group.
Tissue expression pattern analysis indicated that CaEPSPS was constitutively expressed in leaves, stems and roots, with the lower expression being found in roots. The coding sequence
of CaEPSPS gene was successfully subcloned in a plasmid-Escherichia coli system (pET-32a), and the cells containing the plasmid carrying the CaEPSPS gene exhibited enhanced tolerance to herbicide glyphosate, compared to the control. 相似文献

A new full-length cDNA encoding 3-hydroxy-3-methylglutaryl-CoA synthase (designated as TmHMGS, GenBank Accession No. AY644708), which catalyses the condensation of acetyl CoA and acetoacetyl CoA to form 3-hydroxy-3-methylglutaryl-CoA
as an early step in the taxol biosynthetic pathway, was isolated from young leaves of Taxus × media by rapid amplification of cDNA ends (RACE) for the first time. The full-length cDNA of TmHMGS contained a 1431 bp open reading frame (ORF) encoding a deduced protein of 476 amino acid residues. The deduced protein had
an isoelectric point of 5.23 and a calculated molecular mass of about 53 kDa. Amino acid sequence comparison analysis showed
that TmHMGS had high similarity with a number of HMGSs ranging from Schizosaccharomyces pombe to humans, with much higher identity with other HMGSs from plants than those from yeast and humans. Phylogenic analysis showed
that TmHMGS had closest relationship with HMGS from Pinus sylvestris. Tissue expression pattern analysis showed that TmHMGS expressed in needles and stems at similar level, but no expression could be detected in roots. Expression of TmHMGS was all induced by under different elicitors such as silver nitrate, ammonium ceric sulphate and methyl jasmonate, revealed

Plant development from microspore-derived embryos in oilseed rape as affected by chilling, desiccation and cotyledon excision 总被引:1,自引:0,他引:1
The present study evaluated the effects of chilling, partial desiccation, cotyledon excision and successive subculture of
microspore-derived embryos on plant development in oilseed rape (Brassica napus L.). The results showed that out of the five media, all the genotypes showed the best response when the embryos were cultured
on the half-strength Murashige and Skoog medium with 2.0 mg dm−3 benzylaminopurine. A cold treatment for 3 or 5 d further increased frequencies of embryo germination (90.0 %) and plantlet
development (58.46 %). Desiccation for one day also increased the embryo germination and plantlet development in all genotypes
tested. Cutting the cotyledons of the embryos at late cotyledonary stage significantly increased the frequency of plantlet
development. The highest rate of plantlet development was obtained from cultures of embryos sampled with size of less than
4.0 mm. The successive subculture further improved the germination and development of plantlets from embryos. In the genotype
ZJU452, the rate of plantlet development reached 99.78 % after the second subculture of embryos. 相似文献
